چکیده

Background The transcription factor MYB46, together with its redundant paralog MYB83, regulates expression of secondary cell wall biosynthesis genes in Arabidopsis [reviewed in 1]. We isolated the Populus MYB46 ortholog, PtxtMYB021 from hybrid aspen (Populus tremula x tremuloides) [2]. Transiently expressed MYB021 transactivated gene promoters of Populus xylan-active enzymes GT43A, GT43B and Xyn10A. Analysis of conserved motifs within these promoters identified the sequence CCACCAAC, which is similar to the AC elements mediating transactivation by MYB transcription factors during lignin biosynthesis, and we showed that this motif is enriched in xylem-specific carbohydrate active enzyme (CAZyme) promoters.

MYB46 modulates disease susceptibility to Botrytis cinerea in Arabidopsis.

In this study, we show that the Arabidopsis (Arabidopsis thaliana) transcription factor MYB46, previously described to regulate secondary cell wall biosynthesis in the vascular tissue of the stem, is pivotal for mediating disease susceptibility to the fungal pathogen Botrytis cinerea. We identified MYB46 by its ability to bind to a new cis-element located in the 5' promoter region of the pathog...

Carbohydrate-active enzymes involved in the secondary cell wall biogenesis in hybrid aspen.

Wood formation is a fundamental biological process with significant economic interest. While lignin biosynthesis is currently relatively well understood, the pathways leading to the synthesis of the key structural carbohydrates in wood fibers remain obscure. We have used a functional genomics approach to identify enzymes involved in carbohydrate biosynthesis and remodeling during xylem developm...

Discovery of fiber-active enzymes in Populus wood

Renewable fibers produced by forest trees provide excellent raw material of high economic value for industrial applications. Despite this, the genes and corresponding enzymes involved in wood fiber biosynthesis in trees are poorly characterized. This thesis describes a functional genomics approach for the identification of carbohydrate-active enzymes involved in secondary cell wall (wood) forma...

Reconstitution of a Secondary Cell Wall in a Secondary Cell Wall-Deficient Arabidopsis Mutant

The secondary cell wall constitutes a rigid frame of cells in plant tissues where rigidity is required. Deposition of the secondary cell wall in fiber cells contributes to the production of wood in woody plants. The secondary cell wall is assembled through co-operative activities of many enzymes, and their gene expression is precisely regulated by a pyramidal cascade of transcription factors. D...

The MYB46/MYB83-mediated transcriptional regulatory programme is a gatekeeper of secondary wall biosynthesis.

BACKGROUND The secondary cell wall is a defining feature of xylem cells and allows them to resist both gravitational forces and the tension forces associated with the transpirational pull on their internal columns of water. Secondary walls also constitute the majority of plant biomass. Formation of secondary walls requires co-ordinated transcriptional regulation of the genes involved in the bio...
